分子背景(Background)
甲型肝炎病毒细胞受体2也称为HAVCR2、FLJ14428、KIM3、TIM3、TIMD3,是调节分子TIM家族的成员,具有一个Ig样V型结构域和富含Ser/Thr的粘蛋白柄。CD4阳性辅助T淋巴细胞根据其细胞因子分泌模式可分为1型(Th1)和2型(Th2)。Th1细胞及其相关细胞因子参与细胞介导的对细胞内病原体的和迟发型超敏反应,而Th2细胞参与控制细胞外蠕虫感染和促进特应性和过敏性疾病。这两种类型的细胞还交叉调节另一种细胞的功能。HAVCR2是一种Th1特异性细胞表面蛋白,调节巨噬细胞活化并增强小鼠实验性自身性脑脊髓炎的严重程度。HAVCR2调节巨噬细胞活化。抑制T辅助性1型淋巴细胞(Th1)介导的自身和同种反应,并促进耐受。也可能参与T细胞归巢。HAVCR2-半乳糖凝集素-9通路的失调可能是人类慢性自身性疾病状态的基础,如多发性硬化症。
